The Effect of Catecholarmines and EDTA on the Na-K-ATPase Activity in Rat Cerebral Cortex

Abstract
|
|
|
The effect of catecholamines, EDTA and divalent metal ions on the Na-K-ATPase activity in rat cerebral homogenate have been investigated.
The results were as follows
1. Noradrenaline, dopamine, isoproterenol and adrenaline stimulated the Na-K-ATPase activity in dosedependent manner.
2. EDTA could mimic the effects of catecholamines. and EDTA increased the Na-K-ATPase activity in dose-dependent manner.
3. Catecholamines increased the Na-K-ATPase activity at an EDTA concentration of 10¢¥ M whereas in the absence of EDTA the increase of Na-K-ATPase activity produced by catecholamines decreased. Little increase of Na-K-ATPase activity produced by catecholamine occurred at an EDTA concentration of 10-3 M.
4. Ca?¢¥, Zn2+ and Fe"¢¥ inhibited the Na-K-ATPase activity. Dopamine at 10¢¥ M and noradrenaline at 10-4 M were completely effective in reversing the inhibitory effect of Fe"¢¥ but ineffective in reversing the inhibitory effect of Cat+ and Zn".
5. Effects of noradrenaline on Fe" inhibition of Na-K-ATPase activity were only partial in reversing the inhibition of enzyme activity by phentolamine and propranolol.
